Perhaps. Sometimes after four or five less serious comments, the overall total seems to cry for a response.
Personally, I think it's very revealing that your main concern is not people joking about rape, but people killing your buzz by saying rape jokes are not funny.

So you have a really good reason then?
I don't think anything should ever be off limits. I think it's fine to point out I didn't find something funny because it seems to be making light of rape or be a joke based on false racial stereotypes or whatever, but I think it is important for us as a species to be able to laugh at our own tragedies.
